    Diving behaviour of whale sharks in relation to a predictable food pulse

    We present diving data for four whale sharks in relation to a predictable food pulse (reef fish spawn) and an analysis of the longest continuous fine-resolution diving record for a planktivorous shark. Fine-resolution pressure data from a recovered pop-up archival satellite tag deployed for 206 days on a whale shark were analysed using the fast Fourier Transform method for frequency domain analysis of time-series. The results demonstrated that a free-ranging whale shark displays ultradian, diel and circa-lunar rhythmicity of diving behaviour. Whale sharks dive to over 979.5 m and can tolerate a temperature range of 26.4 degrees C. The whale sharks made primarily diurnal deep dives and remained in relatively shallow waters at night. Whale shark diving patterns are influenced by a seasonally predictable food source, with shallower dives made during fish spawning periods

    Development, characteristics, and effects of the new Chatham Harbor inlet

    A new tidal inlet into Chatham Harbor, Massachusetts, has developed from a breach in the barrier beach, Nauset Beach, that forms the outer shoreline of southeastern Cape Cod. Increased tidal range and wave energy resulting from the new inlet produced acute coastal erosion and channel shoaling within Chatham Harbor, with significant impacts on the fishing and boating industries, and on private and public propeny and interests. Study results are consistent with the hypothesis that the Nauset-Monomoy barrier beach system undergoes a long-term cycle of geomorphological change, and that a new cycle was initiated with the formation of this new inlet.     Micromanipulation of InP lasers with optoelectronic tweezers for integration on a photonic platform

    The integration of light sources on a photonic platform is a key aspect of the fabrication of self-contained photonic circuits with a small footprint that does not have a definitive solution yet. Several approaches are being actively researched for this purpose. In this work we propose optoelectronic tweezers for the manipulation and integration of light sources on a photonic platform and report the positional and angular accuracy of the micromanipulation of standard Fabry-Pérot InP semiconductor laser die. These lasers are over three orders of magnitude bigger in volume than any previously assembled with optofluidic techniques and the fact that they are industry standard lasers makes them significantly more useful than previously assembled microdisk lasers. We measure the accuracy to be 2.5 ± 1.4 ”m and 1.4 ± 0.4° and conclude that optoelectronic tweezers are a promising technique for the micromanipulation and integration of optoelectronic components in general and semiconductor lasers in particular

    Change in Nutritional Status Modulates the Abundance of Critical Pre-initiation Intermediate Complexes During Translation Initiation \u3cem\u3ein Vivo\u3c/em\u3e

    In eukaryotic translation initiation, eIF2∙GTP–Met-tRNAiMet ternary complex (TC) interacts with eIF3–eIF1–eIF5 complex to form the multifactor complex (MFC), while eIF2∙GDP associates with eIF2B for guanine nucleotide exchange. Gcn2p phosphorylates eIF2 to inhibit eIF2B. Here we evaluate the abundance of eIFs and their pre-initiation intermediate complexes in gcn2 deletion mutant grown under different conditions. We show that ribosomes are three times as abundant as eIF1, eIF2 and eIF5, while eIF3 is half as abundant as the latter three and hence, the limiting component in MFC formation. By quantitative immunoprecipitation, we estimate that ∌ 15% of the cellular eIF2 is found in TC during rapid growth in a complex rich medium. Most of the TC is found in MFC, and important, ∌ 40% of the total eIF2 is associated with eIF5 but lacks tRNAiMet. When the gcn2Δ mutant grows less rapidly in a defined complete medium, TC abundance increases threefold without altering the abundance of each individual factor. Interestingly, the TC increase is suppressed by eIF5 overexpression and Gcn2p expression. Thus, eIF2B-catalyzed TC formation appears to be fine-tuned by eIF2 phosphorylation and the novel eIF2/eIF5 complex lacking tRNAiMet

    Mycobacterium tuberculosis RNA Expression Patterns in Sputum Bacteria Indicate Secreted Esx Factors Contributing to Growth are Highly Expressed in Active Disease

    To identify factors contributing to the ability of tubercle bacilli to grow in the lung during active infection, we analyzed RNA expression patterns in bacteria present in patient sputum. Prominent among bacterial transcripts identified were those encoding secreted peptides of the Esat-6 subfamily that includes EsxK and EsxL (Rv1197 and Rv1198). H37Rv esxKL and esxJI transcripts were differentially expressed under different growth conditions, and disruption of these genes altered growth phase kinetics in typical laboratory batch broth cultures. These growth defects, including the reduced intracellular growth of an ΔesxKL mutant in primary human macrophages, were reversed by either low multiplicity co-infection or co-culture with wild-type bacteria, demonstrating the ability of the secreted factors to rescue isogenic mutants. Complementing either only esxL or esxI alone (Rv1198 or Rv1037c) also reduced observed growth defects, indicating these genes encode factors capable of contributing to growth. Our studies indicate that the Mycobacterium tuberculosis Mtb9.9 family secreted factors EsxL and EsxI can act in trans to modulate growth of intracellular bacteria, and are highly expressed during active human lung infection

    The Signature of Primordial Grain Growth in the Polarized Light of the AU Mic Debris Disk

    We have used the Hubble Space Telescope/ACS coronagraph to make polarization maps of the AU Mic debris disk. The fractional linear polarization rises monotonically from about 0.05 to 0.4 between 20 and 80 AU. The polarization is perpendicular to the disk, indicating that the scattered light originates from micron sized grains in an optically thin disk. Disk models, which simultaneously fit the surface brightness and polarization, show that the inner disk (< 40-50 AU) is depleted of micron-sized dust by a factor of more than 300, which means that the disk is collision dominated. The grains have high maximum linear polarization and strong forward scattering. Spherical grains composed of conventional materials cannot reproduce these optical properties. A Mie/Maxwell-Garnett analysis implicates highly porous (91-94%) particles. In the inner Solar System, porous particles form in cometary dust, where the sublimation of ices leaves a "bird's nest" of refractory organic and silicate material. In AU Mic, the grain porosity may be primordial, because the dust "birth ring" lies beyond the ice sublimation point. The observed porosities span the range of values implied by laboratory studies of particle coagulation by ballistic cluster-cluster aggregation. To avoid compactification, the upper size limit for the parent bodies is in the decimeter range, in agreement with theoretical predictions based on collisional lifetime arguments. Consequently, AU Mic may exhibit the signature of the primordial agglomeration process whereby interstellar grains first assembled to form macroscopic objects.Comment: 12 pages, 8 figures, ApJ, in pres
